Beriso Amane finished second with 02:21:50 as Lonah Salpeter managed a time of 02:21:57 to take third position.
Ababel Yeshaneh and Emma Bates were fourth and fifth. The duo clocked a time of 02:22:00 and 02:22:10 respectively.
Obiri made her marathon debut last year when she ran at the New York City Marathon where she placed sixth.
Her win comes after Evans Chebet successfully defended his Boston Marathon title on April 17, after clocking a time of 2: 05: 54.
Chebet was defending his win in the men's Marathon having won it last year.
Geoffrey Mutai holds the Boston Marathon course record of 2:03:02 from 2011.
